چکیده
The roles of cyclic AMP and cyclic AMP-dependent protein kinase in the contractile and metabolic responses to neural stimulation were studied in the bullfrog heart perfused in situ. The interaction of parasympathetic and sympathetic nerves in the regulation of myocardial cyclic AMP, cyclic AMP-dependent protein kinase, and phosphorylase also was studied. Stimulation of the left vagosympathetic trunk in the presence of atropine (10 JUM), i.e., sympathetic stimulation, caused frequency-dependent increases in ventricular contractility, cyclic AMP concentration, and activities of cyclic AMP-dependent protein kinase and phosphorylase. Maximal responses occurred with stimulation at frequencies of 5-20 Hz. Responses at 5 Hz persisted throughout 1 minute of continuous stimulation. In contrast, at 100 Hz, all responses except phosphorylase activation were transient during 1 minute of continuous stimulation. The frequencyand time-dependence of the cyclic AMP and cyclic AMP-dependent protein kinase responses were found to be very similar to those of the contractile response. These data are consistent with the hypothesis that cyclic AMP and cyclic AMP-dependent protein kinase serve as mediators of the sympathetic contractile response. All biochemical and mechanical responses to sympathetic neural stimulation were blocked by propranolol (1 JUM), suggesting that only /?-adrenergic receptors are involved in the sympathetic responses. In the absence of atropine, stimulation of the left vagosympathetic trunk had no effect on cyclic AMP concentration or the activities of cyclic AMP-dependent protein kinase and phosphorylase. Therefore, simultaneous parasympathetic stimulation totally blocked the sympathetic biochemical responses. This correlates with a total blockade of the sympathetic contractile response by simultaneous parasympathetic stimulation. Stimulation of the left vagosympathetic trunk in the presence of propranolol (without atropine), i.e., parasympathetic stimulation alone, had no significant effect on cyclic AMP concentration or the activities of cyclic AMP-dependent protein kinase and phosphorylase. متن کاملRole of phosphorylase kinase and cyclic AMP-dependent protein kinase in the regulation of phosphorylase phosphatase.
The enzymes of glycogen metabolism are regulated by interconversion between phosphorylated and dephosphorylated forms. In contrast with our well-defined knowledge of the phosphorylation processes of different enzymes and their role in glycogen breakdown and synthesis, the phosphatases catalysing the reverse processes are only now being understood. The regulation of dephosphorylation reactions i...
متن کاملMechanisms of activation of cardiac glycogen phosphorylase in ischemia and anoxia.
The effects of ischemia and anoxia on cardiac adenosine 3',5'-monophosphate (cyclic AMP) concentration, glycogen phosphorylase activity ratio (— 5'-AMP: + 5'-AMP), phosphorylase kinase activity ratio (pH 6.8:8.2), and myocardial contractility (left ventricular dP/dt) were studied in an open-chest rat heart preparation. Ischemia produced by termination of coronary blood flow increased cyclic AMP...

Cyclic GMP-dependent protein kinase, purified to homogeneity from bovine lung, was shown to activate hormone-sensitive lipase partially purified from chicken adipose tissue. The degree of activation was the same as that effected by cyclic AMP-dependent protein kinase although higher concentrations of the cyclic GMP-dependent enzyme were required (relative activities expressed in terms of histon...
متن کاملRegulation of phosphorylase kinase in rat ventricular myocardium. Role of calmodulin.
Conversion of phosphorylase b to a which is catalyzed by the enzyme phosphorylase kinase is known to require Ca++. Trifluoperazine, an inhibitor of calmodulin-dependent enzymes, was utilized in the present study to clarify the role in vivo of calcium-calmodulin regulation of phosphorylase kinase. Twenty-minute preperfusion of isolated rat ventricles with 10(-5) M trifluoperazine had no effect o...
